Regarding the wrist and thumb, have a look at how you're gripping the handle. Don't get a death-grip going on the thing. Your four fingers should be "hooking" the handle without gripping hard, and your thumb can go to the movies for all the work it does. You can let your fingers relax and even wiggle them about during the recovery phase (when you're moving the seat back up to the next catch).

P.S. I have developed carpal tunnel recently and now row with a right wrist brace. That seems to help a lot.
